چکیده

Rutacridone, 1-hydroxy-A-methylacridone, and l-hydroxy-3-methoxy-A-methylacridone have been isolated for the first time from callus cultures, which had been obtained from meristematic stem cells of Ruta graveolens L. The alkaloids were identified by co-chromatography with authentic compounds, by IR spectra, and by electronic spectra before and after addition of A1C13 . The use of the bathochromic shift by the AlCl3-complex in the analysis of acridine-alkaloids has been demonstrated for the first time.

Biosynthesis of Acridone Alkaloids. A Cell-Free System from Ruta graveolens Cell Suspension Cultures

Ruta graveolens, Suspension Cultures, Cell-Free System, Acridone Biosynthesis, 1,3-DihydroxyN-methylacridone A cell-free extract of Ruta graveolens cell suspension cultures, strain R-20, was prepared which synthesizes 1,3-dihydroxy-N-methylacridone from N-methylanthranilic acid and malonylcoenzyme A. The reaction is strongly dependent on the presence of ATP. After adding of coenzyme A to the as...

Purification and cDNA cloning of anthranilate synthase from Ruta graveolens: modes of expression and properties of native and recombinant enzymes.

Ruta graveolens utilizes anthranilate synthase (AS) for the synthesis both of tryptophan in primary metabolism and acridone alkaloids in secondary metabolism. AS has been purified from plants and cell cultures of R. graveolens 670- and 1700-fold, respectively. Glutamine- and ammonia-dependent AS activities were strictly co-purified in all steps. Through cDNA cloning and complementation of Esche...

Genetic transformation of the medicinal plant Ruta graveolens L. by an Agrobacterium tumefaciens-mediated method

Ruta graveolens L. is a source of pharmacologically active molecules such as furocoumarins, furoquinolines and acridone alkaloids. A genetic transformation method using Agrobacterium tumefaciens was developed for this plant species. The conditions for an efficient regeneration of R. graveolens led us to retain hypocotyls among other explants tested. Subsequently, transformation was obtained by ...

Rutacridone-epoxide, a New Acridone Alkaloid from Rut a graveolens *

Several acridone alkaloids have been isolated from roots of Ruta graveolens (for summary see [1, 2]). One of the main alkaloids is rutacridone (1) which seems to be a precursor of several derivatives with hydroxyl or chlorine functions at the isopropyl moiety. As an intermediate an 18,19-epoxide of rutacridone could be postulated [1, 3] that has not yet been identified. Screening for antimicrob...

First syntheses of two quinoline alkaloids from the medicinal herb Ruta chalepensis via cyclization of an o-iodoaniline with an acetylenic sulfone.

The first syntheses of two quinoline alkaloids found in the medicinal herb Ruta chalepensis are reported via the conjugate addition of an o-iodoaniline to an acetylenic sulfone, followed by Pd-catalyzed carbonylation, intramolecular acylation of the corresponding sulfone-stabilized carbanion, and reductive desulfonylation.
